To minimize the risk of gastrointestinal side effects, start dosing at 100 mg 2–3 times daily and slowly increase to effective dose over a period of 2 weeks. Use for at least 1 week to see beneficial effects. Consult a health care practitioner for use beyond 1 year. Relief of symptoms of fibromyalgia: 1 caplet 3–4 times daily with food or as directed by a health care practitioner. To minimize the risk of gastrointestinal side effects, start dosing at 100 mg 2–3 times daily and slowly increase to effective dose over a period of 2 weeks. Cautions:

Serotonin is one of the major neurotransmitters in the body that plays important roles in emotional well-being, sleep, pain, and appetite. Low serotonin can be an underlying factor in multiple chronic health problems.

Supplemental 5-HTP readily crosses the blood-brain barrier and is converted directly into serotonin by the body, providing a direct route to relieving symptoms of low serotonin levels (Birdsall, 1998).
Clinical research confirms that 5-HTP is more successful at alleviating sadness than taking a placebo (Shaw, 2002). A randomized double-blind study supplemented clinically sad patients with 150–400 mg of 5-HTP per day. Within the first two weeks, patient scores on the Hamilton Rating Scale for Depression (HAM-D) significantly decreased. After eight weeks, 73% of the patients showed significant improvements
in emotional wellbeing (Jangid et al., 2013).

A meta-analysis of 13 published trials further supports 5-HTP
as an effective therapy for clinically sad patients. Doses of 50–3250 mg of 5-HTP per day were associated with improved symptoms in 65% of patients. Supplementation also helped lower average HAM-D scores by 12.7 points, moving patients from “severe” sadness to within the “healthy” range (Javelle et al., 2020).

5-HTP can help improve serotonin imbalances associated with chronic pain conditions, such as the musculoskeletal pain, tenderness, morning stiffness, and fatigue experienced by fibromyalgia patients (Birdsall, 1998). Studies lasting up to one year show that fibromyalgia patients who take 300–400 mg of 5-HTP per day in divided doses can significantly improve the severity of symptoms (Birdsall, 1998). In an open study, fibromyalgia patients were supplemented with 100 mg of 5-HTP three times per day. Assessments conducted after 15, 30, 60, and 90 days revealed improved symptoms, with 50% of patients showing a “good” or “fair” level of improvement by the end of the study (Sarzi, 1992).

Migraines can also cause debilitating pain. A double-blind, placebo- controlled study found that 400 mg of 5-HTP per day helped to reduce the frequency and severity of headaches in migraine patients more effectively than a placebo. After two months of supplementation, 52% of patients had fewer headache symptoms (Benedittis et al., 1986).

5-HTP is used as a sleep aid in people with disrupted sleep. In a placebo-controlled study, eight healthy participants took either 200 mg or 600 mg of 5-HTP per day for 17 days. Researchers concluded
that both doses successfully increased the duration of REM sleep in participants. In addition, those taking the 600 mg dose experienced more rapid eye movement (REM) activity (Wyatt et al., 1971).

Normal serotonin levels help the brain recognize feelings of satiety after eating. By increasing serotonin production, 5-HTP can be used to control appetite and carbohydrate cravings. In a placebo-controlled study, obese women with body mass index (BMI) of 30–40 were supplemented with 8 mg/kg of body weight per day for five weeks. Even without having additional dietary restrictions, the women were found to eat less and lose weight during the study (Ceci et al., 1989).

Timed-release caplets deliver 5-HTP gradually, helping to smooth
out the fluctuations in serotonin levels that can occur with immediate release forms. This method of supplementation is especially important for people with poor emotional well-being who cannot risk a relapse of symptoms or adverse events related to rapid spikes in serotonin levels (Jacobsen et al., 2019).
